UCSF study points to potential strategy for erasing memory of addiction
A new study by researchers at the Ernest Gallo Clinic and Research Center at UC San Francisco offers encouraging findings that researchers hope may one day lead to a treatment option for people who suffer from alcohol abuse disorders and other addictions.
In the study, conducted in rats, the UCSF researchers were able to identify and deactivate a brain pathway linked to the memories that cause cravings for alcohol, thus preventing the animals from seeking alcohol and drinking it, the equivalent of relapse.
"One of the main causes of relapse is craving, triggered by the memory by certain cues - like going into a bar, or the smell or taste of alcohol," said lead author Segev Barak, PhD, at the time a postdoctoral fellow in the lab of co-senior author Dorit Ron, PhD, a Gallo Center investigator and UCSF professor of neurology.
"We learned that when rats were exposed to the smell or taste of alcohol there was a small window of opportunity to target the area of the brain that reconsolidates the memory of the craving for alcohol and to weaken or even erase the memory, and thus the craving" he said.
The study, also supervised by co-senior author Patricia H. Janak, a Gallo Center investigator and UCSF professor of neurology, will be published online on June 23, 2013 in Nature Neuroscience.
In the first phase of the study, rats had the choice to freely drink water or alcohol over the course of seven weeks, and during this time developed a high preference for alcohol. In the next phase, they had the opportunity to access alcohol for one hour a day, which they learned to do by pressing a lever. They were then put through a 10-day period of abstinence from alcohol.
Following this period, the animals were exposed for 5 minutes to just the smell and taste of alcohol, which cued them to remember how much they liked drinking it. The researchers then scanned the animals' brains, and identified the neural mechanism responsible for the reactivation of the memory of the alcohol - a molecular pathway mediated by an enzyme known as mammalian target of rapamycin complex 1 (mTORC1).
They found that just a small drop of alcohol presented to the rats turned on the mTORC1 pathway specifically in a select region of the amygdala, a structure linked to emotional reactions and withdrawal from alcohol, and cortical regions involved in memory processing.
They further showed that once mTORC1 was activated, the alcohol-memory stabilized (reconsolidated) and the rats relapsed on the following days, meaning in this case, that they started again to push the lever to dispense more alcohol.
"The smell and taste of alcohol were such strong cues that we could target the memory specifically without impacting other memories, such as a craving for sugar," said Barak, who added that the Ron research group has been doing brain studies for many years and has never seen such a robust and specific activation in the brain.
In the next part of the study, the researchers set out to see if they could prevent the reconsolidation of the memory of alcohol by inhibiting mTORC1, thus preventing relapse. When mTORC1 was inactivated using a drug called rapamycin, administered immediately after the exposure to the cue (smell, taste), there was no relapse to alcohol-seeking the next day. Strikingly, drinking remained suppressed for up to 14 days, the end point of the study. These results suggest that rapamycin erased the memory of alcohol for a long period, said Ron.
The authors said the study is an important first step, but that more research is needed to determine how mTORC1 contributes to alcohol memory reconsolidation and whether turning off mTORC1 with rapamycin would prevent relapse for more than two weeks.
The authors also said it would be interesting to test if rapamycin, an FDA-approved drug currently used to prevent organ rejection after transplantation, or other mTORC1 inhibitors that are currently being developed in pharmaceutical companies, would prevent relapse in human alcoholics.
"One of the main problems in alcohol abuse disorders is relapse, and current treatment options are very limited." Barak said. "Even after detoxification and a period of rehabilitation, 70 to 80 percent of patients will relapse in the first several years. It is really thrilling that we were able to completely erase the memory of alcohol and prevent relapse in these animals. This could be a revolution in treatment approaches for addiction, in terms of erasing unwanted memories and thereby manipulating the brain triggers that are so problematic for people with addictions."

Everything you want to know about Android's new Navigation Drawer
Google's Android developers have delivered a great video all about the new Navigation Drawer
We've talked before about how the Android team is serious about design. They want Android, and the great apps that developers create, to not only work well, but to look good. We noticed this at Google I/O in May, that there was a new (and long-awaited) focus on building applications that look like the belong. Apps with a consistent and elegant user interface, versus the jumble of different design styles and choices of the past.
It's a big undertaking. You have to account for different screen sizes, devices with or without the recommended button layout, and most of all you need to find a fluid look and feel that works well for the end user. Nobody likes an app that is difficult to use. Google tried to get everyone on-board with the action bar method, and it's a great solution for some things. But sometimes, you need to get dirty and have a list of user choices and options. That's where the new Navigation Drawer concept comes into play.
Grab your phone. Open the YouTube app or Google Play Books app. Swipe your finger to the right on the main screen (view). That's the new Navigation Drawer at work. Google Play Books could probably get away with just an action bar and overflow area (those three dots in the upper right) but the YouTube app has plenty of choices and options that would otherwise be buried three levels deep. The Navigation Drawer seems like a perfect way to present them all to the users -- you and me.
A lot of thought went into this, and luckily the Android developers are willing to share their thoughts about it all. Join Roman Nurik, Jens Nagel and Rich Fulcher after the break for 47 minutes of great information about why, where and when you would want to use the Navigation Drawer. Even if you're never going to design an Android app, it's a great watch and really drives the idea home.

?Snowden managed to get away because Hong Kong decided to stall,? adds Claudia Mo, a lawmaker with the pro-democracy Civic Party. ?The matter was too tricky for Sino-American relations ? so Beijing gave instructions he should be given time to leave.?
Snowden had said he planned to challenge any US extradition attempt in Hong Kong courts, declaring his faith in the city?s rule of law. But he faced the possibility of having to stay in jail throughout the court proceedings, which could have taken several years according to local lawyers.
His case was a thorny one for Beijing, anxious to improve relations with the United States and embarrassed by the US fugitive?s presence in Hong Kong, but unable to intervene openly in Hong Kong?s judicial process under the ?one country, two systems? principle that safeguards Hong Kong?s courts.
Hong Kong?s top official, Chief Executive Leung Chun-ying had promised that the case would be handled ?in accordance with the laws and established procedures of Hong Kong.? But the politically sensitive case ?would have been quite a test for our rule of law,? says Ms. Mo. ?It would have been a very thorny issue and it is all for the best for both Hong Kong and Beijing that he has gone.?
?This was not a case that Hong Kong or Beijing ever wanted to get involved in,? agrees Mr. Egan. ?The best thing for both of them was for Snowden to leave.??

Kerry: Political solution urgently needed in Syria
DOHA, Qatar (AP) ? U.S. Secretary of State John Kerry called on Saturday for an urgent political resolution to the war in Syria, saying that unless the bloodshed stops, the region could descend into a chaotic sectarian conflict.
Kerry met in Doha with 10 of his counterparts from Arab and European nations to coordinate aid to the embattled rebels trying to oust Syrian President Bashar Assad in a two-year civil war that has left 93,000 dead. All the nations in attendance agreed to step up aid to the rebels, Kerry said.
While he offered no specifics, Kerry said the assistance would help change the balance on the battlefield, where regime forces have scored recent victories. Kerry blamed Assad for the deteriorating situation in Syria, saying that while the international community was attempting to hold a conference to set up a transitional government, Assad invited Iranian and Hezbollah fighters to bolster his troops.
It was Kerry's first meeting with his counterparts about aid to the Syrian rebels since President Barack Obama announced that the U.S. would send lethal aid to the opposition despite concern that weapons could fall into the hands of Islamic extremists in Syria. That decision was partly based on a U.S. intelligence assessment that Assad had used chemical weapons, but Kerry expressed deeper concern about Iran and Hezbollah fighters.
"That is a very, very dangerous development," Kerry said. "Hezbollah is a proxy for Iran. ... Hezbollah in addition to that is a terrorist organization."
Kerry blamed Hezbollah and Assad with thwarting efforts to diffuse sectarian rebels and to negotiate a settlement.
"We're looking at a very dangerous situation," that had transformed "into a much more volatile, potentially explosive situation that could involve the entire region," Kerry said.
The war already has spilled into neighboring countries and is increasingly being fought along sectarian lines, pitting Sunni against Shiite Muslims and threatening the stability of Syria's neighbors.
Kerry met with his counterparts in the Qatari capital on the first stop of a seven-nation trip through the Mideast and Asia where he is tackling difficult foreign policy issues ? from finding peace between the Israelis and Palestinians to trying to gain traction on U.S. talks with the Taliban to end the Afghanistan war. James Dobbins, U.S. special envoy to Afghanistan and Pakistan, arrived in Doha on Saturday, but talks with the Taliban have not been scheduled.
Kerry seemed to put the ball in the Taliban's court, saying the Americans and Qataris were all on board to help negotiate a political resolution to the war and it was up to the Taliban to come to the table at a new political office they opened last week in Doha. "We are waiting to find out whether the Taliban will respond, Kerry said.
"We will see if we can get back on track. I don't know whether that's possible or not," Kerry said. "If there is not a decision made by the Taliban to move forward in short order, then we may have to consider whether the office has to be closed."
On Syria, Kerry has been pressing hard on Russia to back an international conference intended to end the bloodshed in Syria and allow a transitional government to move the country beyond civil war.
Russia has been the key ally of Assad's regime throughout the two-year conflict.
Top U.S. diplomats are ready to go to Geneva to meet with Russian Foreign Minister Sergey Lavrov and other officials next week to advance the political process, Kerry said. The date and location of the international conference on Syria haven't been announced, but it's already being dubbed "Geneva 2" because a similar event was held there a year ago.
On Friday, Russia's foreign minister said Washington was sending contradictory signals on Syria that could derail an international conference intended to end that country's civil war, warning that U.S. talk about a possible no-fly zone would only encourage the rebels to keep fighting.
Sergey Lavrov also criticized demands that Assad step down. Russian leaders warn that if Assad steps aside, the resulting power vacuum could be quickly filled by al-Qaida connected rebels, who are well-armed and aggressive.

Hawke and Delpy remain as charming as ever, and their combined goofiness is more endearing than annoying.
Love is messy here, life cannot be controlled, satisfaction is far from guaranteed. Romance is rocky at best. But romance still is.
Though "Before Midnight" is often uncomfortable to watch, it's never less than mesmerizing - and ultimately, a joy to walk with this prickly but fascinating couple again.
"Before Midnight" is heartbreaking, but not because of Jesse and Celine. It's the filmmakers' passions that seem to have cooled.
Before Midnight is fascinating to watch, and so long as Celine and Jesse are communicating, there's still hope.
How (Jesse and Celine) try to rekindle that flame is what drives Midnight, a film that feels so authentic it's like overhearing a conversation you're not sure you should be hearing.
Marvelous. It's impossible to shake the feeling that we are merely eavesdropping on reality. Witty, wise, and -- most important of all -- truly romantic in ways that movies usually aren't.
It's been 18 years since Hawke, Delpy and Linklater introduced us to Jesse and Celine, and their story just gets richer, funnier and more punchy each time we see them. In 1995's Before Sunrise, they were idealistic 23-year-olds.
Hawke and Delpy are as believably real as any screen couple can ever be.
This is one of the few sequels for which the cliche 'eagerly awaited' is truly applicable.
Predictably, it's just as great as the first two.
By the end, Before Midnight inches towards a dawn of charm. But it's a troubled trip.
As an organic experiment in collaboration between actors and director, it is a triumph, co-created and co-owned by Delpy, Linklater and Hawke.
Hawke and Delpy, who are both credited on the script too, have never found co-stars to bounce off more nimbly or bring out richer nuances in their acting.
The performances and dialogue are wonderfully naturalistic; a reminder that the best special effects are often the cheapest.
Before Midnight is about the nature of long-term relationships, and the way love deepens and grows but also finds itself subject to the complications of maturity. Smart, insightful, and poignant.
For those who witnessed Jesse and Celine's tentative getting together as inter railing students almost twenty years ago, it's reassuring to see them still in love.
Brilliantly directed, superbly written and impeccably acted, this is a thoroughly enjoyable, thought-provoking and emotionally engaging drama that perfectly complements the previous two films.
It remains as engaging, illuminating, honest and funny as its predecessor; here's hoping we revisit Jesse and C?line in another decade or so.
Nine-year gaps between films would sink a studio franchise, but the unforgiving impact of time and the slipperiness of its mysterious mental record, memory, are the very subjects of 'Before Midnight.'
Contains more shrewd and candid insights than many Hollywood midlife crisis dramas.
A couple persistently digging at each other might have been insufferable to watch except that Hawke and Delpy manage to be goofy, funny, witty and charming, even while they're fighting.

SOFIA (Reuters) - Bulgaria will not impede a push to put Hezbollah's armed wing on the European Union's terrorism blacklist because the new government, after wavering on the issue, has now decided to fall into line with its fellow EU members, diplomats said.
A Black Sea state of 7 million people, Bulgaria was thrust to the front of Middle Eastern diplomacy when the previous government blamed Hezbollah for a bombing that killed five Israeli tourists and a driver in a Bulgarian resort last year.
The accusation played a central part in a Western diplomatic push to blacklist the Lebanon-based group, an effort which has acquired new urgency since its fighters started helping government forces in Syria's civil war.
Bulgaria alarmed many of its EU partners this month when Foreign Minister Kristian Vigenin, from the new Socialist government, backed away from that stance and questioned whether there was enough proof of Hezbollah's role.
However, since then Bulgaria has been lobbied by other EU states and has decided to change direction again, according to two Western diplomats who spoke on condition of anonymity.
"It seems that the new government has been testing its limits and has tried to see how far it may go, but once they got a reaction they quickly stepped back," one diplomat said.
The second diplomat said: "Vigenin has now stated publicly that the position of Bulgaria is unchanged. I believe the government will stick to this position."
Hezbollah has denied any involvement in the bus bombing.

Blacklisting the group - a step that would freeze its assets in Europe and prevent it from raising funds there - would mark an about-turn for the EU, which has long resisted pressure from Washington and Israel to do so.
The debate within the bloc is finely balanced, with some members concerned about the risk of further fuelling tensions in the Middle East. Diplomats say a majority of the 27 states back a British push to put Hezbollah on the list, but unanimity is needed for a decision.
At a meeting in Brussels on Wednesday, Britain failed to win over skeptical delegates, and may now escalate the issue to a higher level, diplomats said.
Officials in the Bulgarian government have rejected suggestions that they made any U-turns over Hezbollah. The foreign ministry said the minister would not speak to Reuters about the issue.
In a statement sent to Reuters on Wednesday, the ministry said: "Bulgaria is ready to join a consensus decision of the European Union. The responsibility to present a solid foundation for it is a collective one."
The Socialist government, while broadly pro-Western, is traditionally sympathetic towards Russia, and this could have played a part, said a former official with the previous, center-right government.
"The new government do not want to irritate the Russians by showing an active position on Hezbollah," said the source, who did not want to be identified.
Moscow, an ally of Syrian President Bashar al-Assad, has blocked international moves to condemn Hezbollah for getting involved in Syria.
Diplomats say there is no evidence the Kremlin directly influenced the Socialist government's policy on Hezbollah, but Russia is a consideration for all Bulgarian leaders.
Bulgaria imports most of its gas from Russia's Gazprom. Moscow is seeking compensation of about 900 million euros from Bulgaria after it cancelled a new nuclear plant that Russian state firm Atomstroyexport had been contracted to build.

Depardieu fined $5,300 in drunk-driving case
PARIS (AP) ? A French court has fined Gerard Depardieu 4,000 euros ($5,300) and suspended his driver's license for six months in a drunk-driving case that started when the actor fell off his scooter in Paris in November and ended up being taken to a police station.
Depardieu's lawyer, Eric de Caumont, said he planned to appeal Friday's ruling. He said Depardieu, who was not at the hearing, was in Moscow filming.
It's not the first time alcohol and the law mixed for the 64-year-old actor. In 1998, Depardieu crashed his motorcycle when his blood-alcohol level was five times over the legal limit. He escaped with leg and face injuries.
Depardieu has publicly feuded with France's ruling Socialist government and accepted a Russian passport from President Vladimir Putin.

Property insurance bill moves forward | Under The Dome
A bill that adds transparency to the state's ratemaking process for homeowners insurance is headed to the House floor after sailing through the House Insurance Committee.
The bill, HB 519, was unanimously approved by a voice vote of committee members after changes were once again made to accommodate issues raised by the Insurance Department and the industry.
"We have addressed several of the concerns," said Rep. Paul Tine, a Democrat from Kitty Hawk and one of the primary sponsors of the bill. "I won't say that everything has been addressed."
The bill requires the N.C. Rate Bureau, which represents insurers on rate issues, to use at least two computer models to calculate potential losses from catastrophes if computer models are used at all. Critics along the coast, where homeowners been hit by much higher rate increases than the rest of the state, have complained about the models used by the industry.
The bill also calls for each industry rate request to include historical data dating back to 1987, broken down by territory, for losses stemming from hurricanes.
An earlier version of the bill called for insurance companies to include in their premium notices to consumers a breakdown of the amount of the premium that stems from insuring against wind and hail damage and how much is related to coverage for fire, theft and other claims.
But the version approved Tuesday calls for the state Insurance Department to post that information for each territory on its website instead.

US employers add 165K jobs; rate falls to 7.5 pct.

WASHINGTON (AP) ? The U.S. economy showed last month why it remains the envy of industrialized nations: In the face of tax increases and federal spending cuts, employers added a solid 165,000 jobs in April ? and far more in February and March than anyone thought.
The job growth in April drove down the unemployment rate to a four-year low of 7.5 percent and sent a reassuring sign that the U.S. job market is improving.
In its report Friday, the Labor Department revised up its estimate of job gains in February and March by a combined 114,000. It now says employers added 332,000 jobs in February and 138,000 in March. The economy has created an average of 208,000 jobs a month from November through April ? above the 138,000 added in the previous six months.
"This is a good report," said John Silvia, chief economist at Wells Fargo. "There's a lot of strength... It's good for the economy. It's good for people's income."
The stronger job growth suggests that the federal budget cutting "does not mean recession," Silvia said. "It does not mean a dramatic slowdown."
Stock prices soared when trading began on Wall Street at 9:30 a.m. Eastern time. The Dow was up 155 points in early trading.
The unemployment rate has fallen 0.4 percentage point since the start of the year, though it remains high. The Federal Reserve has said it plans to keep short-term interest rates at record lows at least until unemployment falls to 6.5 percent.
The hiring last month was broad-based. The only sectors of the economy that cut jobs were construction and government.
Some higher-paying sectors added workers. Professional and technical services, which includes accounting, engineering and architecture, added 23,000 jobs. Education and health services added 44,000.
But some of the bigger job gains were in lower-paying fields, such as hotels and restaurants, which added 45,000 jobs, and retail, which added 29,000. Temporary help firms gained 31,000 positions.
The job growth is occurring while the U.S. economy is growing modestly but steadily. It expanded at a 2.5 percent annual rate in the January-March quarter, fueled by the strongest consumer spending in two years.
The global economy, by contrast, is slowing. The European Union warned Friday, for example, that the 17 countries that use the euro currency will shrink by a collective 0.4 percent this year. And unemployment across the eurozone is expected to hit an average of 12.2 percent. In Greece and Spain, it's forecast to reach 27 percent.
Both Fed Chairman Ben Bernanke and European Central Bank President Mario Draghi have suggested that governments need to focus on stimulating growth and not just on spending cuts and deficit reduction.
In April, more Americans said they had part-time jobs even though they wanted full-time work. That figure rose 278,000 to 7.9 million, reversing a steep drop the previous month.
Some economists worry that restaurants, retail chains and other companies are hiring more part-time workers in preparation for the implementation of health care reform. Companies with more than 50 full-time employees in 2013 will be required to provide health insurance to their full-time staff next year.
The revisions to the March and February figures were unusually large. Retailers, restaurants and hotels added 48,000 more jobs in February than previously reported. They accounted for three-quarters of that month's revision.
The government revises each month's job totals twice in the following two months. The revisions occur because many companies in the survey submit their responses late.
The average workweek for private-sector employees declined 0.2 hour to 34.4 hours, but average hourly earnings rose 4 cents to $23.87. In the past year, wages have risen faster than inflation.
The number of people who have been unemployed for more than six months dropped 258,000 to 4.4 million. Over the past year, the number of long-term unemployed has declined by 687,000.

NEW YORK (AP) ? Barnes & Noble is teaming up with Google to vastly increase the number of apps available on its Nook HD tablets.
The bookstore chain says it will add Google's Play app store to its 7-inch Nook HD and 9-inch HD+ products in the U.S. and U.K. via a software update Friday. The move expands the number of apps available from the roughly 10,000 the Nook already offered in its own store ? such as Angry Birds and Netflix ? to 700,000-plus apps and games offered on Google Play. And it comes after a weak holiday sales season for the Nook, which is struggling to gain market share in the rapidly expanding tablet market.
CEO William Lynch said research and sales during the holidays show that consumer preference is shifting toward all-purpose tablets rather than simple e-readers.
"We saw coming off holiday the market moved to multifunction tablets," he said. "Consumer research showed us the breadth of applications available is really critical."
Lynch said the company had been in discussions with Google "on and off" for the past several years.
"This addresses the one perceived gap that we had with other tablets virtually overnight," he said. Terms of the deal were undisclosed.
The update is automatic and will occur over-the-air to all devices connected to Wi-Fi. It will also include other Google Inc. services like the Chrome browser, Gmail, YouTube and Google Maps. Google Play Music includes millions of songs as well.
The prices and styles of the Nooks that Barnes & Noble offers are not changing. The 7-inch Nook HD starts at $199 and the 9-inch Nook HD+ tablet starts at $269. Barnes & Noble also sells non-tablet e-book readers, the Nook Simple Touch and Nook Simple Touch with GlowLight, which will not offer Google Play.
Barnes & Noble Inc., based in New York, has invested heavily in its Nook e-book readers and a digital library as more readers shift to electronic books and competition has grown from discount stores and online rivals.
The retailer launched high-definition versions of its Nook HD and Nook HD+ tablets in September in an effort to better compete against Amazon.com's Kindle Fire as well as other tablets like the iPad, iPad Mini and Google's own Nexus 7.
The company's Nook unit has attracted investors ? Microsoft Corp. owns 16.8 percent, while U.K. publisher and education company Pearson PLC has a 5 percent stake. But aside from investor funding, it has not been profitable. In its most recent quarter, which included the holiday season, Nook revenue fell 26 percent to $316 million. Barnes & Noble recorded $21 million in returns due to weak demand during the holiday season, and $15 million in allowances for promotions.
Meanwhile market share has slid. Barnes & Noble had a 1.9 percent share of the worldwide tablet market in the fourth quarter, making it the No. 5 Tablet player behind Apple, Samsung, Amazon and Asus, according to data from IDC. But by the first quarter it had slipped out of the top 5, replaced by Microsoft.
At the same time, competition is proliferating, with the global tablet market growing quickly. It more than doubled to 49.2 million units during the first quarter, according to IDC's estimate.

Possible lead in case of missing U.S. journalist in Syria
BOSTON (AP) ? The New Hampshire-based family of a journalist missing for five months now believe "with a very high degree of confidence" that he is being held in a Syrian prison.
James Foley was last seen on Nov. 22 in northwest Syria, where he was contributing videos to Agence France-Press for the media company GlobalPost. His family in Rochester, N.H., says he was kidnapped by unknown gunmen.
GlobalPost CEO Philip Balboni said Friday that an exhaustive investigation has determined that Foley was likely abducted by a pro-Syrian government military group. Investigators believe he is being held with one or more Western journalists in a detention facility near Damascus.
Foley's family and the company have appealed to the Syrian government to release him.
The family spoke at World Press Freedom Day in Boston.
